| dc.description.abstract | Para la obtención de ésteres alquílicos por transesterificación de aceites vegetales se han empleado catalizadores químicos que requieren condiciones fuertes de operación, poseen un alto poder de corrosión y presentan dificultades en la recuperación del glicerol y la remoción del catalizador. Las lipasas pueden ser utilizadas como biocatalizadores, estas trabajan en condiciones suaves y medios libres de solventes, son biodegradables y su reutilización es posible si son inmovilizadas. En este trabajo se llevó a cabo la obtención de esteres etílicos a partir del aceite de palmiste, empleando como catalizador lipasa inmovilizada de Thermomyces lanuginosus. La primera fase del desarrollo experimental consistió en la determinación del peso molecular promedio de un triacilglicerol en el aceite y seguidamente la evaluación de los efectos de difusión interna y externa en el sistema, posteriormente se seleccionaron las mejores condiciones de reacción aplicando un diseño experimental de tres variables por tres niveles, basado en un diseño central compuesto rotatorio de 23 + estrella, analizado por metodología de superficie de respuesta con ayuda del software estadístico Statgraphics centurión XV. Finalmente, se realizó la evaluación de la estabilidad operativa de la enzima y la cinética de la reacción. Se encontró que no se presentan restricciones de difusión interna ni externa en el sistema y que la variable más influyente sobre el porcentaje de conversión es la cantidad de enzima mientras que la agitación lo es sobre el índice de acidez. Las condiciones de reacción seleccionadas (Relación de sustratos 4:1-alcohol:aceite, 9% de enzima con respecto al peso del aceite, 100 rpm y 40 º C) permiten alcanzar conversiones de 96,7 ± 1,6 % con un índice de acidez de 1,3 ± 0,1. La formación de acilgliceroles parciales es mínima (1,47%) y la enzima presenta buena estabilidad operativa. | |
| dc.description.abstractenglish | For the obtaining of alkyl esters by transesterification of vegetal oils, chemical catalysis have been used, this process requires strong operation conditions, and has high corrosion power. It also presents difficulties in the recovery of glycerol and removal of the catalyst. Lipases can be used as biocatalyst, they work in smooth conditions and free solvent mediums, they are biodegradable and its reusability is possible if they are immobilized. In this work, it was carried out the obtaining of ethyl esters from palm kernel oil, using lipase immobilized of Thermomyces lanuginosus as catalyst. First stage of the experimental development was the determination of the average molecular weight of one triacylglycerol in the oil and next, the evaluation of internal and external diffusion effects in the system. Later, the best reaction conditions were selected by applying an experimental design of three variables by three levels, based on a central design compound of 23 + star, it was analyzed by response surface methodology with aid of the statistical software, Statgraphics centurión XV. Finally, it was evaluated the operative stability of the enzyme and the kinetic of the reaction. It was found that there are not restrictions of internal nor external diffusion in the system and that the variable that is most influent on the percentage of conversion is the amount of enzyme whereas the agitation is it on the total acid number. The conditions of reaction selected (Substrates relation 4:1- alcohol:oil, 9 % of enzyme with respect to weight of the oil, 100 rpm and 40 º C) allow to reach 96.7 ± 1.6% of conversions with a total acid number of 1.3 ± 0,1.
