dc.description.abstract | El cáncer de pulmón es la principal causa por muerte de cáncer en el mundo, dado que tiene opciones limitadas de tratamiento y generalmente se detecta en un estado avanzado. Por tanto, el estudio y desarrollo de compuestos efectivos contra esta enfermedad se convierte en una ardua tarea para la comunidad científica. Estudios anteriores sobre el compuesto sintético tipo tetrahidroquinolinaisoxazol, THQIZJ3, mostraron actividad citotóxica sobre las células tumorales de pulmón, sin afectar las células sanas. Por esta razón, con el fin de evaluar la interacción del compuesto THQIZJ3 en la línea celular A549 de cáncer de pulmón y conocer las posibles alteraciones metabólicas generadas, se realizó un análisis por cromatografía de gases acoplado a espectrometría de masas y posterior análisis estadístico de los datos por métodos uni- y multivariados. Metabolitos involucrados en el ciclo de Krebs, glucólisis y ruta de las pentosas fosfato fueron tentativamente identificados y relacionados con cambios metabólicos ocurridos posterior al tratamiento. La alteración de las rutas metabólicas identificadas en el estudio, están estrechamente relacionadas con la producción de energía a nivel celular, por cuanto permite explicar la inhibición en la proliferación celular generado en la línea celular A549. A pesar de que estudios adicionales son necesarios para fundamentar y correlacionar los perfiles químicos de la línea celular de cáncer de pulmón aquí reportados, con su nivel de expresión de los genes y proteínas implicadas en las vías metabólicas identificadas como afectadas, es importante resaltar que el estudio abre un panorama interesante para proponer este tipo de compuestos químicos como moléculas promisorias en la búsqueda de nuevas alternativas quimioterapéuticas para enfrentar el cáncer de pulmón. | |
dc.description.abstractenglish | Lung cancer is the world's leading cause of cancer death, as it has limited treatment options and is usually detected at an advanced stage. Therefore, the study and development of effective compounds against this disease becomes an arduous task for the scientific community. Previous studies on the synthetic tetrahydroquinolineisoxazole compound, THQIZJ3, showed cytotoxic activity on lung tumor cells, without affecting healthy cells. For this reason, to evaluate the interaction of the compound THQIZJ3 in the A549 lung cancer cell line and to unravel the possible metabolic alterations generated, a metabolomic approach based on gas chromatography coupled to mass spectrometry analysis with subsequent statistical analysis of the data by univariate and multivariate methods was carried out. Metabolites involved in the Krebs cycle, glycolysis and the pentose phosphate pathway were tentatively identified and related to metabolic changes that occurred after treatment. The alteration of the metabolic pathways identified in the study are closely related to the production of energy at the cellular level, since it allows to explain the inhibition on cell proliferation generated in the A549 cell line. Although additional studies are necessary to substantiate and correlate the chemical profiles of the lung cancer cell line reported here, with their level of gene expression and proteins involved in the metabolic pathways identified as affected, it is important to highlight that this study opens up an interesting outlook to propose this type of chemical compounds as promising molecules in the search for new chemotherapeutic alternatives to face lung cancer. | |
