dc.description.abstract | El polimorfismo es un campo relevante en la industria farmacéutica ya que puede afectar las propiedades en el estado sólido de un fármaco, por lo cual estudiar este fenómeno permite el desarrollo de nuevos medicamentos mediante la ingeniería de cristales. El objetivo de este trabajo fue realizar un estudio polimórfico de la Nitazoxanida, de los diferentes productos obtenidos en procesos de cristalización, utilizando difracción de rayos X de monocristal para su determinación estructural y posterior análisis de solubilidad en el equilibrio. El material obtenido en el ensayo con la mezcla acetona: agua (E2NTZ), cristaliza en un sistema monoclínico con parámetros de celda a=66,9564(16) Å, b=5,36334(17) Å, c=14,5446(5) Å, α=β=γ=90°, siendo una estructura pseudosimétrica debido a que las relaciones entre los parámetros de la celda unidad corresponden a un sistema ortorrómbico pero las operaciones de simetría corresponden a un sistema monoclínico. El sólido obtenido por la evaporación lenta de solvente en metanol (E4NTZ), cristaliza en un sistema ortorrómbico con parámetros de celda a=14,5542(2) Å, b=5,3959(1) Å, c=33,4777(4) Å, α=β=γ=90°, similar a la reportada en la base de datos CSD para el polimorfo I, indicando la transformación de fase durante este proceso. La determinación estructural del E2NTZ y E4NTZ permite establecer que la Nitazoxanida puede cristalizar en más de una forma, presentando la nueva fase una solubilidad 1,3 veces mayor. | |
dc.description.abstractenglish | Polymorphism is of great interest in the pharmaceutical industry because it can affect the properties of a drug in the solid state. Studying this phenomenon allows the development of new drugs through crystal engineering. The aim of this work was to conduct a polymorphic study of Nitazoxanide, of the different products obtained through crystallization processes, using monocrystalline X-ray diffraction for structural determination and their subsequent equilibrium solubility analysis. The material obtained in the test with the acetone mixture: water (E2NTZ), crystallizes in a monoclinic system with cell parameters a=66,9564(16) Å, b=5,36334(17) Å, c=14,5446(5) Å, α=β=γ=90°, being a pseudo-symmetric structure because the relationships between the unit parameters correspond to an orthorhombic system but the symmetry operations correspond to a monoclinic system. The solid obtained by the slow evaporation of solvent in methanol (E4NTZ), crystallizes in an orthorhombic system with cell parameters a=14,5542(2) Å, b=5,3959(1) Å, c=33,4777(4) Å, α=β=γ=90°, similar to the data reported in the CSD database for polymorph I, indicating the phase transformation during this process. The structural determination of E2NTZ and E4NTZ allows us to establish that Nitazoxanide can crystallize in more than one form, presenting the new phase a solubility 1.3 times higher. | |
