dc.description.abstract | En el presente trabajo se buscó sintetizar y caracterizar un hidrogel basado en alginato y óxido de grafeno mediante gelificación iónica, para encapsular células beta pancreáticas BRIN BD11, con el fin de simular un ambiente apropiado para el crecimiento y proliferación celular, con potencial aplicación para el tratamiento de diabetes tipo 1. Se estudió la influencia del GO a tres concentraciones diferentes (10, 25 y 50 µg/mL) dentro de la matriz de alginato (ALGO10, ALGO25 y ALGO50). En el proceso de caracterización se emplearon diferentes tecnologías para analizar las propiedades fisicoquímicas, mecánicas y biológicas de los hidrogeles. Los resultados de este estudio mostraron a través del SEM que al aumentar el contenido de GO en los hidrogeles, se presentó mayor rigidez y grosor en la superficie, con uniformidad de las hojuelas de GO, evidenciando la correcta incorporación del mismo a la matriz de alginato. Adicionalmente, se determinó que los hidrogeles ALGO25 y ALGO50 presentaron mayor entrecruzamiento, dando como resultado una estructura más compacta, menos hinchada y con menor tasa de degradación. Por otro lado, se observó a través del DMA que al adicionarle GO hubo un aumento significativo en el módulo de elasticidad para los hidrogeles ALGO25 y ALGO50 (0.0055 y 0.0056 MPa), esto es debido a que se formaron enlaces que reforzaron la matriz polimérica, presentando un comportamiento elástico similar al del páncreas. En cuanto a las propiedades biológicas se presentaron resultados favorables con el aumento del contenido de GO, ya que su viabilidad fue de 143,61 y 199,67% para los hidrogeles ALGO25 y ALGO50, respectivamente. Siendo el hidrogel ALGO25 el candidato potencial para la encapsulación celular ya que obtuvo mejores resultados en cuanto a citotoxicidad y proliferación de las células BRIN BD11. Este estudio permite tener un punto de partida para investigaciones futuras en el área de biomateriales. | |
dc.description.abstractenglish | In the present work we sought to synthesize and characterize a hydrogel based on alginate and graphene oxide by ionic gelification, to encapsulate pancreatic beta cells BRIN BD11, in order to simulate an appropriate environment for cell growth and proliferation, with potential application for the treatment of type 1 diabetes. The influence of GO was studied at three different concentrations (10, 25 and 50 µg/mL) within the alginate matrix (ALGO10, ALGO25 and ALGO50). Different technologies were used in the characterization process to analyze the physico-chemical, mechanical and biological properties of the hydrogels. The results of this study showed through the SEM that by increasing the GO content in the hydrogels, greater rigidity and thickness was presented on the surface, with uniformity of the GO leaflets, evidencing the correct incorporation of the same to the alginate matrix, In addition, it was determined that the hydrogels ALGO25 and ALGO50 showed a greater cross-linking, resulting in a more compact structure, less swollen and with a lower rate of degradation. On the other hand, it was observed through the DMA that when adding GO there was a significant increase in the modulus of elasticity for the hydrogels ALGO25 and ALGO50 (0.0055 and 0.0056 MPa), this is because bonds were formed that reinforced the polymer matrix, showing an elastic behavior similar to that of the pancreas. Regarding biological properties, favorable results were presented with the increase in GO content, since its viability was 143.61 and 199.67% for the hydrogels ALGO25 and ALGO50, respectively. The ALGO25 hydrogel was the potential candidate for cell encapsulation as it obtained better results in terms of cytotoxicity and proliferation of BRIN BD11 cells. This study provides a starting point for future research in the area of biomaterials. | |
