Estandarización de marcadores microsatelites heterologos en muestras de vaccinium floribundum del Páramo de Santurban

dc.description.abstract | Vaccinium floribundum Kunth (1819), comúnmente conocido como agraz, mortiño, arándano o blueberry andino, es considerada una especie promisoria por sus características organolépticas, propiedades nutricionales y alto contenido de antioxidantes presentes en el fruto. Las condiciones óptimas para su crecimiento y desarrollo se encuentran en las regiones de páramo y subpáramo. En Colombia, su cultivo comercial se limita a Antioquia y Boyacá, aunque en algunos departamentos como Santander se realiza la cosecha y comercialización del fruto silvestre presentes en zonas de páramo y subpáramo. De manera general, en el país no se han realizado estudios que permitan establecer la diversidad genética de las poblaciones naturales a partir de marcadores moleculares, siendo esta información importante para definir la estructura poblacional y crear planes de uso, manejo y conservación adecuados. Razón por la cual, se estableció como objetivo estandarizar las condiciones de amplificación de marcadores moleculares tipo microsatélite (Simple Secuence Repeats) heterológos en la especie V. floribundum. Los resultados indican que esta especie de arándano es de amplia distribución en el Páramo de Santurbán. A partir del protocolo modificado por Doyle y Doyle (1991) se obtuvo ADN con calidad suficiente para realizar PCR. Se estandarizaron las condiciones para amplificar siete microsatélites heterólogos en V. floribundum, los cuales en general son polimórficos y permitirían detectar diversidad genética en sus poblaciones. Este conocimiento será base para futuros proyectos que permitan determinar la diversidad y estructura de las poblaciones naturales a partir de marcadores moleculares, información necesaria para generar los planes de manejo, uso y conservación de esta especie. | |
dc.description.abstractenglish | Vaccinium floribundum Kunth (1819), commonly known as agraz, mortiño, blueberry or Andean blueberry, is considered a promising species for its organoleptic characteristics, nutritional properties and high content of antioxidants present in the fruit. The optimum conditions for its growth and development are found in the Paramo and Sub paramo regions. In Colombia, its commercial cultivation is limited to Antioquia and Boyacá, although in some departments, such as Santander, the harvest and commercialization of the wild fruit present in Paramo and Sub paramo zones are carried out. No studies have been carried out in the country to establish the genetic diversity of natural populations from molecular markers, this information being important for defining the population structure and creating plans for appropriate use, management and conservation. For this reason, the objective was to standardize the amplification conditions of microsatellite-type molecular markers (Simple Sequence Repeats) in the species V. floribundum. The results indicate that this species of blueberry is widely distributed in the Paramo of Santurbán. Based on the protocol modified by Doyle and Doyle (1991), DNA with enough quality was obtained to carry out PCR. The conditions were standardized to amplify seven heterologous microsatellites in V. floribundum, which are generally polymorphic and would allow the detection of genetic diversity in their populations. This knowledge will be the basis for future projects that allow the determination of the diversity and structure of natural populations based on molecular markers, information needed to generate management plans, use and conservation of this species. | |
