dc.description.abstract | El cáncer de cabeza y cuello (HNC) representa el séptimo cáncer más prevalente en el mundo. Colombia, para 2020, reportó 3.148 nuevos casos y 1343 muertes por esta neoplasia. El 90% de estos tumores son carcinomas de células escamosas (HNSCC), siendo su principal factor de riesgo el consumo de tabaco. El receptor Aril de hidrocarburos (AhR), codificado por el gen AHR, induce la activación de enzimas metabolizadoras de xenobióticos; es posible que diferencias en la actividad de estas enzimas, sean causadas por variantes genéticas sugiriendo que diferencias interindividuales podrían contribuir con susceptibilidad al cáncer. El objetivo fue identificar variantes en el gen AHR en pacientes con HNSCC y su posible efecto sobre la estructura y función de AhR. Este estudio incluyó muestras de sangre de 23 pacientes con HNSCC y mediante secuenciación Sanger, se analizó la región codificante del gen AHR y sus límites intrón-exón. Se determinó la presencia y localización de variantes usando BLAST. Se realizó análisis in silico con herramientas de predicción funcional para establecer el posible efecto de las variantes sobre la estructura y función del receptor. Se reporta para Colombia variantes genéticas en el gen AHR en pacientes con HNSCC. Se identificaron 7 alteraciones de secuencia, de las cuáles 3, no se encuentran reportadas en bases de datos y literatura científica. De las variantes detectadas, una corresponde a la región no traducida 5’ (-459A>G, 5’UTR); 3 de tipo intronicas (c.908+33G>T; c.253+175T>C; c.66-165delT); 2 de región codificante (c.132T>C; c.1661G>A) y una variante de la región no traducida 3’ (c.2333C>T, 3’UTR). Sólo 2 de las variantes identificadas se encuentran reportadas en ClinVar como “benignas”. Este estudio hace un primer acercamiento a la identificación de SNPs en el gen AHR en población colombiana y contribuye al entendimiento de la susceptibilidad genética al cáncer de cabeza y cuello en Colombia. | |
dc.description.abstractenglish | Head and neck cancer (HNC) represents the 7th most common cancer by incidence worldwide. In Colombia, 3.148 new cases and 1343 deaths were reported in 2020 (Sung et al, 2020). The 90% of these tumors are squamous cell carcinomas (HNSCC) and the strongest risk factor for developing HNSCC is tobacco consumption (Johnson et al, 2020). The Aryl hydrocarbon receptor (AhR), encoded by the AHR gene, induces the activation of xenobiotic-metabolizing enzymes. Differences in the activity of these enzymes may be caused by genetic variants in AHR, which suggest the contribution of interindividual differences to cancer susceptibility. We aimed to identify genetic variants in the AHR gene in Colombian smoker patients with HNSCC and their possible effect on the receptor structure and function. The study included blood samples from 23 HNSCC patients. The AHR gene coding sequence and its exon–intron boundaries were sequenced through Sanger’s method. We determined the presence and localization of variants using BLAST. In silico analysis was performed with functional prediction tools to establish the possible effects of the variants on the protein structure and function. We reported for Colombian population genetic variants in the AHR gene in patients with HNSCC. A total of 7 sequence alterations were identified. However, 3 variants are not reported in genomic databases and not found in the scientific literature. The variants detected in AHR corresponded to a 5' prime UTR variant (-459A>G, 5'UTR); 3 intronic type alterations (c.908+33G>T; c.253+175T>C; c.66-165delT); 2 coding region variants (c.132T>C; c.1661G>A) and 1 variant located in the 3' prime UTR region (c.2333C>T, 3'UTR). We found 2 variants reported in ClinVar as "benign". The findings represent a first approach to identify SNPs in the AHR gene in Colombian population. The study contributes to the understanding of genetic susceptibility to HNC in Colombia. | |
