Especificidad y posible mecanismo de acción del péptido antimicrobiano gam019 y sus análogos gam020, gam022 y gam026 contra bacterias gram positivas y gram negativas

dc.description.abstract | Actualmente, los efectos secundarios derivados del uso de antibióticos y la creciente resistencia bacteriana se han convertido en uno de los mayores retos para la salud. Los péptidos antimicrobianos (PAMs) son pequeñas moléculas catiónicas que hacen parte de la solución a este reto; Son generadas por el sistema inmune innato de gran variedad de organismos vivos que abarcan todos los órdenes de vida. Exhiben un amplio espectro de acción, especificidad y distintas conformaciones estructurales lo cual les facilita unirse e interactuar con las membranas celulares bacterianas generando daño interno y en su morfología. En este trabajo se evaluó la actividad y la selectividad o especificidad de cuatro PAMs sintéticos GAM019, GAM020, GAM022 y GAM026, los cuales tienen una corta cadena de 17 aminoácidos (a excepción de GAM026, el cual posee 25 aminoácidos) en su estructura primaria. Se realizaron cambios en la posición de algunos aminoácidos con el fin de variar la estabilidad de su estructura secundaria alfa hélice y evaluar el efecto en bacterias Gram positivas y Gram negativas. Se encontró que estos péptidos actúan en ambos grupos de bacterias y presentaron baja actividad citotóxica en línea celular HepaRG y baja actividad hemolítica en glóbulos rojos. El péptido GAM019 exhibió mayor potencial antibacteriano, con Concentración Mínima Inhibitoria de 15.1 y 26.9 μM para Staphylococcus aureus ATCC 9123 y Escherichia coli ATCC 95222, respectivamente. También se encontró que GAM019 genera cambios en la morfología bacteriana, afecta la respiración bacteriana y promueven la formación de Especies Reactivas de Oxígeno. Estos resultados sugieren que la estructura de alfa hélice es clave para la actividad antimicrobiana que presentan los péptidos. GAM019 es el péptido con mayor porcentaje de helicicidad y a su vez quien presentó actividad antimicrobiana significativa a diferencia de los otros péptidos. Lo anterior, convierte a GAM019 en un buen candidato para ser usado como un antimicrobiano de amplio espectro. | |
dc.description.abstractenglish | Currently, the side effects derived from the use of antibiotics and the growing bacterial resistance have become one of the greatest challenges for health. Antimicrobial peptides (PAMs) are small cationic molecules that are part of the solution to this challenge; They are generated by the innate immune system of a great variety of living organisms that cover all orders of life. They exhibit a wide spectrum of action, specificity, and different structural conformations, which makes it easier for them to join and interact with bacterial cell membranes, generating internal damage and in their morphology. In this work, the activity and selectivity or specificity of four synthetic PAMs GAM019, GAM020, GAM022 and GAM026, which have a short chain of 17 amino acids (with the exception of GAM026, which has 25 amino acids) in their primary structure, were evaluated. Changes were made in the position of some amino acids in order to vary the stability of their alpha helix secondary structure and to evaluate the effect on Gram positive and Gram-negative bacteria. These peptides were found to act on both groups of bacteria and showed low cytotoxic activity in HepaRG cell line and low hemolytic activity in red blood cells. The GAM019 peptide exhibited the highest antibacterial potential, with a Minimum Inhibitory Concentration of 15.1 and 26.9 μM for Staphylococcus aureus ATCC 9123 and Escherichia coli ATCC 95222, respectively. It was also found that GAM019 generates changes in bacterial morphology, affects bacterial respiration and promotes the formation of Reactive Oxygen Species. These results suggest that the alpha helix structure is key to the antimicrobial activity of peptides. GAM019 is the peptide with the highest percentage of helicicity and, in turn, the one that presented significant antimicrobial activity unlike the other peptides. This makes GAM019 a good candidate for use as a broad-spectrum antimicrobial. | |
