Análisis funcional in silico de RNAs largos no codificantes (lncRNAs) de astrocitos humanos bajo estrés lipotóxico y su asociación con enfermedades neurodegenerativas

dc.description.abstract | Las enfermedades neurodegenerativas (EN) son trastornos progresivos e irreversibles que causan deterioro neurológico gradual. El incremento de factores de riesgo como la obesidad han contribuido al aumento de casos de EN. La obesidad puede derivar en lipotoxicidad, afectando la función de neuronas y astrocitos a través de la inducción de factores neuroinflamatorios y otros efectos que pueden contribuir al desarrollo de las EN. Sin embargo, los mecanismos moleculares implicados en este proceso no están completamente esclarecidos. Recientemente se ha demostrado que los RNA largos no codificantes (lncRNA) cumplen un papel regulador en los astrocitos y en las EN, lo que sugiere que el estudio de estas moléculas puede mejorar el conocimiento sobre los procesos neurodegenerativos. Con base en lo anterior, el objetivo de este trabajo fue explorar el rol funcional de lncRNAs asociados a lipotoxicidad. Para ello, se seleccionaron dos lncRNA con expresión diferencial, obtenidos de astrocitos bajo condiciones lipotóxicas usando palmitato y bajo condiciones basales. Estos lncRNAs se analizaron usando DIANA-LncBase y DIANA-TarBase para establecer sus interacciones con miRNAs y mRNAs. Las redes de interacción resultantes se visualizaron empleando Cytoscape y el papel de las redes ceRNA en rutas metabólicas se dilucidaron a través de PANTHER y DIANA-miRpath. Las redes obtenidas se enriquecieron en procesos relacionados con las EN. Además, se propusieron 3 posibles ejes lncRNA/miRNA/mRNA basados en el perfil transcriptómico que podrían involucrarse en los mecanismos de las EN. Los tres ejes, AC092687.3/hsa-let-7e-5p/SOD2, AC092687.3/hsa-let-7e-5p/PMAIP1 y SERTAD4-AS1/hsa-miR-125b-5p/NUDT16L1 mostraron importancia en la regulación de procesos de apoptosis e inflamación, características relevantes de las EN. Estas redes y ejes propuestos ayudarán a comprender los mecanismos inducidos por la lipotoxicidad en astrocitos, ampliar el conocimiento sobre los lncRNAs y su papel regulador en las EN, identificando así moléculas potencialmente útiles para su diagnóstico temprano e implementación de terapias efectivas para contrarrestarlas. | |
dc.description.abstractenglish | Neurodegenerative diseases (ND) are progressive and irreversible disorders that cause gradual neurological deterioration. Increasing risk factors such as obesity have contributed to the increase in cases of ND. Obesity can lead to lipotoxicity, affecting the function of neurons and astrocytes through the induction of neuroinflammatory factors and other effects that may contribute to the development of ND. However, the molecular mechanisms involved in this process are not fully elucidated. Recently, long non-coding RNAs (lncRNAs) have been shown to play a regulatory role in astrocytes and in ND, suggesting that the study of these molecules may improve our understanding of neurodegenerative processes. Based on the above, the aim of this work was to explore the functional role of lncRNAs associated with lipotoxicity. For this purpose, two differentially expressed lncRNAs were selected, obtained from astrocytes under lipotoxic conditions using palmitate and under basal conditions. These lncRNAs were analyzed using DIANA-LncBase and DIANA-TarBase to establish their interactions with miRNAs and mRNAs. The resulting interaction networks were visualized using Cytoscape and the role of ceRNA networks in metabolic pathways was elucidated using PANTHER and DIANA-miRpath. The obtained networks were enriched in ND-related processes. In addition, 3 possible lncRNA/miRNA/mRNA axes were proposed based on transcriptomic profiling that could be involved in ND mechanisms. The three axes, AC092687.3/hsa-let-7e-5p/SOD2, AC092687.3//hsa-let-7e-5p/PMAIP1 and SERTAD4-AS1/hsa-miR-125b-5p/NUDT16L1 showed importance in the regulation of apoptosis and inflammation processes, relevant features of ND. These proposed networks and axes will help to understand the mechanisms induced by lipotoxicity in astrocytes, expand the knowledge about lncRNAs and their regulatory role in ND, thus identifying potentially useful molecules for their early diagnosis and implementation of effective therapies to counteract them. | |
